We are recruiting an experienced motivated Senior Research Associate to join our growing Immunology – Translational Sciences team. The Immunology team is developing methods to understand AAV biology in large animal models to contribute to nonclinical development of gene therapy products for neurological indications.

Responsibilities

  • Develop, perform and analyze AAV and transgene immunogenicity assays in various matrices using plate-based immunoassay methods
  • Design and conduct plate-based ELISAs, electrochemiluminescence (ECL)- based (e.g., MSD platform) and/or bead-based (e.g., Luminex multiplex platform) immunoassays

Requirements

  • Bachelor’s degree in Biological Sciences with 5+ years of experience or Master’s degree in Biological Sciences with 3+ years of experience in the pharmaceutical industry
  • Background knowledge in immunology, neuroscience or gene-based therapy concepts preferred
  • Strong practical knowledge of protein immunogenicity method development, cut-point calculations and data analysis
  • Familiarity with concepts of method development, qualification and validation in accordance with regulatory guidelines is preferred

Encoded is seeking a Senior Scientist of Translational and Nonclincial Bioanalytical Sciences to perform and oversee the nonclinical biodistribution and dose-determining biochemical studies in the development of gene therapy products. The successful candidate will be a scientific leader that can expand our assays and methods to develop our understanding of in vivo vector biodistribution and transgene expression profile for gene therapy product and contribute to interpretation of nonclinical pharmacology and toxicology studies to support clinical development programs.

Ideal candidate will be enthusiastic, entrepreneurial scientist motivated to shape the development of gene therapy products. As an important member of the Translation research team, this individual will be responsible for method development, sample testing, data analysis and integration of results for proof-of-concept and pivotal non-clinical studies leading to regulatory filings. Strong analytical and method development skills with ability to perform and coordinate activities both within internal teams (research groups) and external partners (large animal CROs) are required.

Responsibilities:

  • Contributes to the development and execution of bioanalytical studies for candidate gene therapy products, throughout the drug development lifecycle
  • Provide high quality analytical assessments for tissue biodistribution and mRNA expression profiles with sensitive quantitative molecular biology tools like qPCR, digital droplet (dd)PCR, RT-ddPCR and other pertinent platform technologies.
  • Familiar with sensitive and robust protein detection methods to detect and quantify tissue transgene protein expression profiles using biochemical methods like electrochemiluminescence (ECL)-based Immune assays on MSD platform, Luminex, mass spectrometry approaches and other technologies.
  • Provide molecular biology expertise in the analysis and interpretation of results to integrate with the overall nonclinical studies
  • Provide scientific monitoring and guidance on outsourcing studies for method transfer, troubleshooting, qualification and validation studies to support gene therapy development
  • Author and review method development SOPs, qualification, validation and sample testing reports for regulatory submissions
  • Succinctly and effectively communicate interpretations of nonclinical molecular biodistribution and biochemical studies at cross-functional meetings
  • Stay updated with knowledge of regulatory agency nonclinical and gene therapy regulations/guidelines, and relevant scientific literature

Requirements

  • PhD in Biochemistry, Molecular Biology or equivalent
  • 5+ years experience including industry including postdoctoral work in fast-paced environments
  • Depth of experience in AAV gene therapy or related genetic therapies is essential.
  • Broad knowledge and background in central nervous systems is a plus.
  • Extensive knowledge in conducting molecular assays for DNA and RNA measurements with qPCR, digital droplet PCR, Reverse transcription-PCR methods in a variety of biological matrices. Encoded is looking for an experienced and highly motivated Scientific Study Manager who will be responsible for centrally managing activities required for the effective design, execution, and completion of outsourced Good Laboratory Practices (GLP) and non-GLP primate studies to assess safety and vector biodistribution for CNS indications. The successful candidate will be an engaged member of the Translational team that will proactively manage multiple aspects of external studies, including, but not limited to, study planning and design, protocol development, study coordination and monitoring, study execution, data collection, and reporting activities. For Good Laboratory Practice (GLP)-regulated studies, the position will be responsible for GLP audits and ensuring compliance with appropriate GLP regulations.

Responsibilities:

  • Effectively manage all aspects of outsourced GLP and non-GLP primate studies with minimal supervision, high accuracy, and scientific integrity.
  • Manage progress of studies to ensure adherence to the protocol, objectives, and overall strategy.
  • Anticipate and efficiently resolve/troubleshoot protocol or study-related issues or discrepancies.
  • Monitor the execution of primate studies via on-site visits and frequent interactions via email/video conferencing.
  • Lead efforts to coordinate with Encoded Supply Chain and Biorepository teams to ensure timely and accurate sample shipments between internal and external collaborators.
  • Maintain and organize data files and reports across outsourced studies.
  • Utilize program management skills to develop, track, and ensure study related deliverables are achieved according to scheduled timelines.
  • Travel: Up to 10% domestic travel required to conduct on-site study monitoring as the designated sponsor representative.

Requirements

  • PhD in biology, neuroscience, or related sub-discipline with 6 or more years of pharmaceutical/biotechnology experience in large animal model preclinical toxicology and GLP-testing (21CFR, Part 58) OR
  • Master’s in biology, neuroscience, or related sub-discipline with 10 or more years of pharmaceutical/biotechnology experience in large animal model preclinical toxicology and GLP-testing (21CFR, Part 58)
  • Proven ability to work with a high level of integrity and accuracy, supported by excellent time management and organizational skills.
  • Strong communication and interpersonal skills (oral and written) that enable the individual to interact proactively with CROs, facilitate external meetings, and provide frequent study updates to internal teams.
  • Demonstrated history of working in cross-functional teams supporting safety assessments for biologics in primates.

Encoded is seeking a Director of Safety Toxicology and Pathology to provide scientific leadership for preclinical studies in animal models in the development of cutting-edge gene therapy products for genetic pediatric neurological diseases. This position reports to the VP of Translation Sciences. The successful candidate will be an experienced leader and will serve as a toxicology and pathology expert for nonclinical gene therapy studies in preclinical models. The incumbent will contribute to the design, conduct and interpretation of nonclinical toxicology and pathology assessments for investigational studies in rodent and primate models.

This is an ideal position for a highly enthusiastic, interactive, entrepreneurial individual with demonstrated expertise in toxicology and pathology to shape the development of gene therapy products. Requirements

  • Responsible for the design, qualification and execution of a variety of protein-based analytical methods leading to selection of development candidates.

Requirements

  • Strong working knowledge and technical proficiency in protein biochemistry. Examples include ELISA/MSD-ELISA/western blotting/LICOR based protein quantification, subcellular fractionation, methods to examine/quantify post-translational modifications etc.
  • Demonstrated capability of generating data-driven hypotheses and designing/developing properly controlled studies/assays for hypothesis testing.
  • Experience with molecular biology methods (such as RNA/DNA extractions, qPCR, transfections) and molecular cloning.
  • Experience with biochemistry/molecular biology analysis in cell culture – primary neuronal cell culture or IPSC-derived neuronal cell culture experience is beneficial.

This position will lead a small team of dedicated in vivo researchers supporting the advancement of a variety of projects within the area of monogenic neurological diseases. The Director will work in a collaborative drug discovery environment to bring forward novel gene therapies and will oversee, design and execute in vivo experiments in translational disease models, including transgenic mouse lines, to test efficacy and mechanism of action of novel gene therapy candidates. Key objectives include establishing novel in vivo translational phenotypic assays, demonstrating proof-of-concept data, enabling candidate selection, and generating high quality preclinical pharmacology packages in support of IND-enabling studies.

Responsibilities:

  • Strategic planning and oversight of the in vivo pharmacology evaluation of gene therapies in alignment with departmental pipeline objectives and gene therapy precedents
  • In vivo study design to enable clear go/no-go decision making for therapeutic programs
  • Identification of appropriate Contract Research Organizations for in vivo model execution including negotiating contract details
  • Bring forward innovative ideas, novel translational animal models and advanced technologies to support team goals. Identify opportunities and develop ideas to enhance and accelerate business processes.
  • Work closely with the Senior Director of In Vivo Operations to coordinate efforts across the department, including the development of SOPS and IACUC protocols
  • Responsible for career development of team of diverse scientists, contribute to talent strategy and provide mentorship
  • Contribute to nonclinical component of regulatory submissions; communicate nonclinical pharmacology findings to cross-functional teams with a balanced and clear point of view; prepare for and participate in regulatory agency interactions
  • Cultivate cross-functional relationships with project teams, translational medicine, biomarker teams, and disease-specific working groups to deliver novel targets, biomarkers and drug candidates
  • Achieve pipeline goals by managing resource allocation and oversight of progress across a diverse set of early research programs
  • Present scientific accomplishments internally and externally to the scientific community and discuss with key opinion leaders
  • Support experimental work in the lab if needed
  • Be recognized as a people leader on site to set and maintain a healthy culture.

Requirements

  • Ph.D. degree in pharmacology or neurobiology with at least 8+ years of experience in drug discovery/development in biotech or pharmaceutical industry OR experience performing progressively advanced duties at the Senior Principal Scientist / Associate Director level
  • Background in gene therapy or neurological disorders
  • Proven record in discovery and development of novel therapeutic entities including progression and screening of novel drug candidates and mechanistic studies in preclinical disease models
  • Experience in the development and validation of novel translational disease models
  • Peer reviewed publication record in relevant fields, including participation in preparation of manuscripts patents, pharmacology reports and program reviews
  • Team leadership and people management experience

Preferred Qualifications:

  • Experience in contributing to a disease area strategy creation, development and execution in a Pharma/Biotech setting

Research Associate II/Senior Research Associate, Preclinical Molecular Research

Responsibilities:

  • Independently conduct and troubleshoot in vitro experiments and ex vivo assays in a Preclinical R&D environment with some guidance
  • Perform experiments in mammalian cell culturing with high proficiency
  • Demonstrate a high proficiency in molecular cloning and primer design
  • Demonstrate strong technical capabilities in MCP tasks including but not limited to: DNA/RNA/protein extraction from animal tissue and cultured cells, PCR, qPCR, cDNA library preparations, NGS sequencing, Western blotting, ELISA execution.
  • Analyze and interpret molecular, biochemical, and in vivo data with methodological rigor. Select and run appropriate statistical tests.

Requirements
